4-bromobenzene sulfonamide can be used as intermediates in pharmaceutical synthesis. It can be prepared by the reaction of 4-bromobenzene sulfonyl chloride with ammonia water, and can also be prepared by one-step diazo salt of p-bromobenzene boron tetrafluoride.
4-bromobenzene sulfonamide can be used to synthesize 4- (5-aldehyde – furan) benzene sulfonamide: The compounds 4-bromobenzene sulfonamide (1.0 equivalent), 5-aldehyde -furan-2-boric acid (1 day, 1.5 equivalent), anhydrous sodium carbonate (2.0 equivalent) and bistriphenylphosphine palladium chloride (0.1 equivalent) were dissolved in MeCN:H2O = 2:1, replaced with argon for three times, and heated to 80℃ for 2h. After TLC detection, the reaction was complete. After cooling to room temperature, the insoluble catalyst was filtered out with diatomite and concentrated under reduced pressure. Then, 4- (5-aldehyde – furan) benzene sulfonamide was extracted by ethyl acetate with water and concentrated by column chromatography.
